Abstract:
A tissue fragmenting device and methods of using the same are provided herein. A device for preparing a plurality of tissue fragments from a tissue sample includes a platform having a central opening, a shuttle slidably coupled to the platform and including a shuttle blade, a blade array mounted across the central opening of the platform having a plurality of blades each generally perpendicular to the shuttle blade, and a piston slidably coupled to the platform and slidably coupled to the shuttle along a predefined raceway and configured to urge the tissue sample into the blade array.

CLAIMS

We claim:

1. A device for preparing a plurality of tissue fragments from a tissue sample, comprising: a platform having a central opening; a shuttle slidably coupled to the platform and including a shuttle blade; a plurality of cutting elements mounted across the central opening of the platform and comprising a plurality of blades each generally perpendicular to the shuttle blade; and a piston slidably coupled to the platform and slidably coupled to the shuttle along a predefined raceway and configured to urge the tissue sample into the blade array.

2. The device of claim 1, wherein the platform comprises a channel and the shuttle includes a projection slidably coupled to the platform through the channel.

3. The device of claim 1, wherein the raceway comprises a plurality of races.

4. The device of claim 3, wherein each race of the plurality of races is coupled to an adjacent race by an extending increment ramp.

5. The device of claim 3 or 4, wherein each race of the plurality of races is separated by about 0.5 mm to about 10 mm.

6. The device of claim 5, wherein each race of the plurality of races is separated by about 1 mm to about 5 mm.

7. The device of claim 6, wherein each race of the plurality of races is separated by about 3 mm.

8. The device of claim 1, wherein the plurality of tissue fragments each have a thickness of about 0.5 mm to about 10 mm.

9. The device of claim 8, wherein the plurality of tissue fragments each have a thickness of about 1 mm to about 5 mm.

10. The device of claim 9, wherein the plurality of tissue fragments each have a thickness of about 3 mm.

11. The device of any one of the preceding claims, wherein the shuttle comprises a guide pin slideably received in the raceway.

12. The device of any one of the preceding claims, wherein moving the shuttle through the raceway moves the shuttle blade and then the blade array through a bottom portion of the tissue sample.

13. The device of any one of claims 1 to 11, wherein moving the shuttle through the raceway moves the blade array and then the shuttle blade through a bottom portion of the tissue sample.

14. The device of any one of the preceding claims, further comprising a motor configured to move the shuttle relative to the platform as defined by the shape of the raceway.

15. The device of any one of the preceding claims, wherein the raceway comprises at least two raceways.

16. The device of any one of the preceding claims, wherein the device further comprises a biasing member disposed between the piston and the platform, and wherein the biasing member biases the piston to move towards the platform.

17. The device of claim 16, wherein movement of the shuttle towards the piston actuates the biasing member to move the piston towards the platform to urge the tissue sample into the blade array.

18. The device of claim 17, wherein actuation of the biasing member moves a guide pin in the shuttle slideably received in a race in the raceway up an extending ramp into an adjoining race in the raceway, thereby moving the piston towards the platform to urge the tissue sample into the blade array.

19. The device of any one of claims 1-18, wherein the shuttle blade is double-edged.

20. The device of claim 19, wherein each edge is angled or has a triangular shape, each edge is rectangular or is straight, each edge has a curved shape, or each edge has a crescent shape.

21. The device of any one of claims 1 to 20, wherein the plurality of cutting elements mounted across the central opening is an array of metal blades.

22. The device of any one of claims 1 to 20, wherein the plurality of cutting elements mounted across the central opening is an array of wires.

23. The device of any one of claims 16 to 22, wherein the platform comprises a channel with a first end and a second end, wherein the shuttle includes a projection slidably coupled to the platform through the channel, and wherein the movement of the shuttle from a position proximal to the first end or the second end of the channel towards the piston actuates the biasing member to move the piston towards the platform to urge the tissue sample into the blade array.

The device of claim 23, wherein actuation of the biasing member moves a guide pin in the shuttle slideably received in a race with a first terminus and a second terminus in the raceway up a first extending ramp adjacent to the first terminus or up a second extending ramp adjacent to the second terminus into an adjoining race in the raceway, thereby moving the piston towards the platform to urge the sample into the blade array.

A method of using the device of any one of the preceding claims to cut a tissue sample from a patient into a plurality of tissue fragments, the method comprising loading the tissue sample into a tissue sample mount and moving the shuttle through the raceway to cut the tissue sample in two axes to prepare a plurality of tissue fragments from the tissue sample.

26. The method of claim 25, wherein the plurality of tissue fragments each have a thickness of about 0.5 mm to about 10 mm.

27. The method of claim 26, wherein the plurality of tissue fragments each have a thickness of about 1 mm to about 5 mm.

28. The method of claim 26, wherein the plurality of tissue fragments each have a thickness of about 3 mm.

29. The method of claim 26, wherein the plurality of tissue fragments each have a thickness of about 1 mm.

30. The method of any one of claims 25 to 29, wherein the plurality of tissue fragments each have a width of about 0.5 mm to about 10 mm.

31. The method of any one of claims 25 to 30, wherein the plurality of tissue fragments each have a width of about 1 mm to about 5 mm.

32. The method of any one of claims 25 to 30, wherein the plurality of tissue fragments each have a width of about 3 mm.

33. The method of any one of claims 25 to 30, wherein the plurality of tissue fragments each have a width of about 1 mm.

34. The method of any one of claims 25 to 33, further comprising placing the plurality of tissue fragments into one or more tissue culturing devices.

35. The method of any one of claims 25 to 33, further comprising cryopreserving the plurality of tissue fragments.

36. The method of any one of claims 25 to 33, further comprising storing the plurality of tissue fragments.

CROSS REFERENCE TO RELATED APPLICATIONS

[000] This application claims the benefit of U.S. Provisional Application Serial No. 63/299,346, filed January 13, 2022, which is incorporated herein by reference in its entirety.

FIELD OF THE DISCLOSURE

[001] The disclosure described herein relates generally to a device for reducing tissue samples to fragments and more particularly, but not exclusively, to a plurality of uniform fragments.

BACKGROUND OF THE DISCLOSURE

[002] A need in the field exists for dividing or fragmenting tissue samples of varied composition, density, and properties into uniform fragments for use in biochemical assays and procedures.

SUMMARY OF THE DISCLOSURE

[003] The devices provided herein meet the needs in the field by providing tissue fragmentation devices capable of cutting tissue samples in at least two axes to provide uniform tissue fragments.

[004] In an embodiment, the disclosure includes a device for preparing a plurality of tissue fragments from a tissue sample. In some embodiments, the tissue sample is a tumor sample obtained from a subject (e.g., a human or animal subject). In some embodiments, the device includes a platform having a central opening. In some embodiments, the device includes a shuttle slidably coupled to the platform and including a shuttle blade. In some embodiments, the device includes a blade array mounted across the central opening of the platform and comprising a plurality of blades each generally perpendicular to the shuttle blade. In some embodiments, the device includes a piston slidably coupled to the platform and slidably coupled to the shuttle along a predefined raceway and configured to urge the tissue sample into the blade array. In some embodiments, moving the shuttle through the raceway moves the shuttle blade and the blade array through a bottom portion of the tissue sample to cut the plurality of tissue fragments from the tissue sample.

[005] In some embodiments, the platform comprises a channel and the shuttle is slidably coupled to the platform through the channel.

[006] In some embodiments, the raceway comprises a plurality of races. In some embodiments, each race of the plurality of races comprises an increment ramp. In some embodiments, each race of the plurality of races is separated by about 0.5 mm to about 10 mm. In some embodiments, each race of the plurality of races is separated about 1 mm to about 5 mm. In some embodiments, each race of the plurality of races is separated by about 3 mm. In some embodiments, each race of the plurality of races is separated by about 1 mm.

[007] In some embodiments, each tissue fragment that may be prepared by the device described herein may be of uniform size. In some embodiments, the plurality of tissue fragments may be uniform. In some embodiments, the plurality of tissue fragments have a thickness of about 0.5 mm to about 10 mm. In some embodiments, the plurality of tissue fragments have a thickness of about 1 mm to about 5 mm. In some embodiments, the plurality of tissue fragments have a thickness of about 3 mm. In some embodiments, the plurality of tissue fragments have a thickness of about 1 mm.

[008] In some embodiments, the shuttle may comprise a guide pin that may be connected to the raceway. In some embodiments, the shuttle comprises at least one guide pin that is slidably connected to the raceway.

[009] In some embodiments, moving the shuttle through the raceway moves the shuttle blade and then the blade array through a bottom portion of the tissue sample. In some embodiments, moving the shuttle through the raceway moves the blade array and the shuttle blade through a bottom portion of the tissue sample.

[0010] In some embodiments, the devices described herein may include a motor configured to move the shuttle through the raceway.

DETAILED DESCRIPTION OF THE DISCLOSURE

[0025] A number of assays are known in the art that may require tissue fragments or pieces of uniform and consistent size. For example, adoptive cell therapy (ACT) relies on the use of tumor fragments or pieces for the culturing and expansion of endogenous tumor reactive T-cells (e.g., Tumor Infiltrating Lymphocytes), which may then be delivered to cancer patients. However, animal tissue, and especially cancerous animal tissue, does not have a uniform density or texture and provides a challenge when attempting to reduce such tissue to uniform fragments or pieces. As used herein, the term “tissue” refers to animal tissue. In some embodiments, the tissue may be tumor tissue. In some embodiments, a tissue sample may be a solid tumor or a portion thereof that may have been surgically excised from a patient. In some embodiments, a tissue sample may be tumor biopsy sample.

[0026] To meet the needs in the field, devices of the disclosure allow for the fragmentation and cutting of a tissue sample into a number of uniform fragments or pieces by cutting the sample in multiple axes.

[0029] Referring now to the figures, wherein like elements are numbered alike throughout, FIGS. 1-9 refer to a device 1 for cutting and fragmenting tissue to provide a plurality of tissue fragments.

[0030] FIGS. 1-3 provide various views of a device 1 that may be used to fragment and cut tissue and thereby provide a plurality of tissue fragments or pieces. In an embodiment, the device 1 includes a platform 100, and shuttle 200, and a piston 300. As shown in FIG. 1, the platform 100, shuttle 200, and piston 400 are movably coupled to one another as described herein. [0031] Platform 100 may include a blade array 400 and may be coupled to the shuttle 200 via channels 110, which may be disposed on either side of the platform 100. In some embodiments, platform 100 may be movably coupled to shuttle 200 through the channels 110. In some embodiments, platform 100 includes one channel 110. In some embodiments, platform 100 includes at least two channels 110.

[0032] In some embodiments, shuttle 200 includes one or more guide pin(s) 220 that register with channels 110 and allow for the shuttle 200 to track in a predetermined sliding configuration relative to the platform 100. For example, the shuttle 200 may be configured to slide horizontally with respect to the platform 100 as guide pin(s) 220 track with the channels 110.

[0033] Shuttle 200 may also include a blade 230 (also referred to herein as “shuttle blade” 230) configured to cut a tissue sample along a horizontal axis (i.e., a Z-axis) (see FIGS. 4A-4B).

Blade 230, for example, may be a ceramic blade or a metal blade (e.g., stainless steel). FIGS.

8 A- 8H illustrate possible blade shapes of some embodiments. In some embodiments, blade 230 includes a single sharpened edge (“cutting edge”) configured to cut through the tissue sample X. In some embodiments, blade 230 includes at least one cutting edge. In other embodiments, as shown in FIG. 5B, the shuttle blade 230 may have two edges, a first edge 231 and a second edge 232. In some embodiments, both first edge 231 and second edge 232 are configured as cutting edges, e.g., sharp enough to cut through the tissue sample. Edges 231 and 232 may be positioned on opposite sides of blade 230. In some such embodiments, for example, having two cutting edges allows blade 230 to cut the tissue sample from either direction as blade 230 is moved back and forth with respect to the tissue sample, as will be described further herein.

[0034] In some embodiments, the blade 230 has a rectangular shape (Fig. 8A) or has a parallelogram shape (Fig. 8D), for example, such that first edge 231 and second edge 232 are straight and substantially parallel. In some embodiments, the blade 230 has a triangular shape, trapezoidal shaped, or is angled (Fig. 8B). In some such embodiments, first edge 231 and second edge 232 may be obliquely angled with respect to each other. In some embodiments, blade 230 has a polygonal shape, for example, a concave polygonal shape (e.g., FIG. 8E) or a convex polygonal shape (e.g., FIG. 8F). In some such embodiments, rather than having a straight linear configuration, edges 231 and 232 may each have one or more angles. In some embodiments, one or more edges of the blade 230 has a curved shape (e.g., FIG. 8C). In some embodiments, the blade 230 has a crescent shape (Figs. 8G and 8H). As noted above, in some embodiments, the shuttle blade 230 is double-edged, for example, where each of edges 231 and 232 is sharpened for slicing through the tissue sample. In some embodiments, the shuttle blade 230 is doubleedged, and each edge is angled or has a triangular shape (Fig. 8E or 8F). In some embodiments, the shuttle blade 230 is double-edged, and each edge is rectangular or is straight (FIG. 8A, 8D). In some embodiments, the shuttle blade 230 is double-edged, and each edge has a curved shape (FIG. 8C). In some embodiments, the shuttle blade 230 is double-edged, and each edge has a crescent shape (FIG. 8G, FIG.8H).

[0035] Figs. 1, 2, 3A-1, and 3A-2 illustrate piston 300 with a tissue mount 310 and a raceway 500. Raceway 500 may have a first end 502 and a second end 503. Raceway 500 may include a plurality of races 510. Races 510 may be disposed in a substantially parallel configuration on one or more sides of piston 300. Races 510 may be defined by longitudinal ribs 512. Longitudinal ribs 512 may be positioned above and below each race 510, for example. Fig. 3 A-2 is illustrated with seventeen longitudinal ribs. As illustrated in Fig. 3 A-2, ribs 512 may include a top longitudinal face 512a and a bottom longitudinal face 512b which may be generally parallel to each other. In some embodiments, top longitudinal face 512a is shorter than bottom longitudinal face 512b. For example, ribs 512 may include a ramped face 512c on one or two lateral ends of rib 512. Ramped face 512c maybe disposed between top longitudinal face 512a and bottom longitudinal face 512b. Ramped face 512c, in some embodiments, extends from top longitudinal face 512a to bottom longintudinal face 512b and may be obliquely angled with respect to top longitudinal face 512a and acutely angled with respect to bottom longintudinal face 512b. Advantages of ramped face 512c includes facilitiating a smooth transition of pin 210 into race 510 from one or both ends of race 510.

[0036] In some embodiments, piston 300 also includes guide(s) 540. In some embodiments, piston 300 includes one or more sets of guides 540. For example, in the embodiment shown in Fig. 3A-1, there is a first set of guides 540 that are separate from and positioned between longitudinal ribs 512 and the first end 502 of raceway 500. In the embodiment shown in Fig. 3 A-2, a second set of guides 540 may be present and positioned between longitudinal ribs and second end 503 of raceway 500. In some embodiments, each set of guides 540 includes a plurality of guides 540 that may be vertically aligned. In some embodiments, the number of guides 540 included in each set may be equal to the number of longitudinal ribs 512. Guides 540, in some embodiments, may be disposed in horizontal alignment with ribs 512. In some embodiments, as illustrated in Figs. 9A-9C, guide(s) 540 have an upper surface 901 and a lower surface 902. The upper and lower surfaces 901 and 902 can be configured to engage guide pin(s) 210 as described in more detail below. In some embodiments, a pair of adjacent guides 540 are separated by a pinway 920 that is defined between the lower surface 902 of a first guide 540 of the pair and the upper surface 901 of the second guide 540 of the pair. In the illustrated embodiment, upper surface 901 includes two longitudinal edges, a first longitudinal edge 907 and a second longitudinal edge 908. First longitudinal edge 907 may be parallel (or substantially parallel) to second longitudinal edge 908. For example, first longitudinal edge 907 and second longitudinal edge 908 may be oriented substantially horizontally when piston 300 is in operation. In some embodiments, in each guide 540 the second longitudinal edge 908 may be vertically offset from first longitudinal edge 907. For example, the second longitudinal edge 908 may be positioned higher than first longitudinal edge 907.

[0037] Upper surface 901 may also include an upper connector 904. In some embodiments, upper connector 904 is interposed between first longitudinal edge 907 and second longitudinal edge 908. In some embodiments, upper connector 904 may include a curved face (e.g., a convexly curved face). The curved face of upper connector 904 may be defined by at least one radius. In some embodiments, upper connector 904 has a curvature with a radius that approximates the radius of the guide pin(s) 210. In some embodiments, the curvature of upper connector 904 is defined by a convex circular arc that extends from first longitudinal edge 907 to second longitudinal edge 908.

[0038] The lower surface 902 of each guide 540 may have two or more planar edges. For example, lower surface 902 may include a first lower planar edge 903 and a second lower planer edge 906. First lower planar edge 903 and second lower planar edge 906 may be non-parallel with respect to one another. In some embodiments, first lower planar edge 903 and second lower planar edge 906 may be configured such that respective projections of first lower planar edge 903 and second lower planar edge 906 intersect within adjacent pinway 920 below lower surface 902. The first lower planar edge 903 may be disposed at an acute angle with respect to the second longitudinal edge 908 of the upper surface 901. The second lower planar edge 906 may be angled acutely in the downward direction with respect to the first longitudinal edge 907 of the upper surface 901. Guide 540 may have an interior end 540b and an exterior end 540a. The predetermined period of time may be, for example, from 1 day to 28 days, from 1 day to 8 days, from 3 days to 11 days, from 3 days to 14 days, from 7 days to 11 days, from 7 days to 14 days, from 11 days to 22 days, etc.

For example, once a tumor sample is introduced into the closed system, the system is not opened to the outside environment until the final population of TILs have been harvested. Transfer of the tissue fragments obtained from device 1 to the tissue culture device(s) and/or transfer of the TILs from one tissue culture device to another may occur, for example, via closed tubing between the components that is sealed from the outside environment. In some embodiments, device 1 and/or the tissue culture devices may be enclosed in an incubator configured to maintain these components at a predetermined temperature (e.g., about 37 °C) and/or atmospheric condition (e.g., 5% CO2).
