Abstract:
Although differentiating pluripotent stem cells by temporarily differentiating the pluripotent stem cells into CTraS considerably increases the subsequent cell differentiation efficiency, it is difficult to stably induce pluripotent stem cells into CTraS in a reproducible manner, and thus, there is a problem in that, consequently, the efficiency of inducing the pluripotent stem cells to differentiate into target neural cells is low. In addition, in the case in which pluripotent stem cells need to be differentiated into target neural cells in the future, it is necessary to differentiate the pluripotent stem cells into the neural cells in a more efficient manner. The inventors of the present invention found a novel method capable of inducing pluripotent stem cells into a state in which the pluripotent stem cells are stable and easily induced to differentiate into neural cells in the future, and thus, demonstrated that the above problem can be solved. Specifically, the inventors demonstrated that the above problem can be solved by culturing cells for five days by adding four types of small molecules, namely, SB431542, LDN193189, SU5402, and EC23.
