The present invention relates to a novel kynureninase and a use thereof for cancer treatment. The novel kynureninase demonstrates similar or higher catalytic activity and thermal stability compared to already known kynureninases, and is more effective in reversing T cell proliferation inhibition induced by L-kynurenine (L-KYN) in an ex vivo culture system. Therefore, the novel kynureninase can be used as a powerful therapeutic enzyme to overcome the immunosuppressive tumor microenvironment. Particularly, the novel kynureninase shows excellent tumor growth inhibition effects in tumor animal models and can be used as an effective agent for various cancer immunotherapies with high industrial applicability.
