Abstract:
This microscope is provided with an excitation light output part, an optical system, and a harmonic detection part. The excitation light output part outputs excitation light. The time waveform of the light intensity of the excitation light includes the nth root (where n is an integer of 2 or more) of a linear function of a sine wave. The maximum value of the light intensity of the excitation light exceeds the saturation excitation intensity in an observed object. The optical system irradiates the observed object with excitation light outputted from the excitation light output part. Fluorescence is produced in the observed object by n-photon excitation due to irradiation of the excitation light. The harmonic detection part detects a second harmonic included in the time waveform of the light intensity of the fluorescence.
