A microfluidic device, comprising a rotating mechanism (1), a rotating assembly (2) and a microfluidic assembly (3), wherein the rotating assembly (2) is rotationally connected to the microfluidic assembly (3) by means of the rotating mechanism (1); the rotating assembly (2) comprises a first limiting mechanism (22), a rotating assembly body (23) and a sealing layer (24), the lower surface of the sealing layer (24) being provided with microfluidic channels (71, 72, 73, 74), which are used for a fluid passing therethrough; and the microfluidic assembly (3) comprises a cover plate (31) and a flow channel plate (32), flow channel ports (41, 42, 43, 44) and liquid via hole pairs (61, 62, 63, 64) being provided on the cover plate (31), a plurality of flow channel segments being provided on the flow channel plate (32), and the liquid via hole pairs (61, 62, 63, 64) being capable of matching with the microfluidic channels (71, 72, 73, 74) on the sealing layer (24) such that the corresponding flow channel segments are in communication.
