Abstract:
The present invention relates to a method for using compartmentalization to increase the yield of 7-dehydrocholesterol in yeast. In the present invention, yeast is used as a starting strain, and heterologous sterol △24-reductase and △-cholestanolase are expressed. The yield of 7-DHC in Saccharomyces cerevisiae S288C is 10.15mg/L. By means of our invention, some enzymes in the 7-DHC synthesis pathway are, by using oxidosomal and mitochondrial positioning tags, positioned in various compartments in Saccharomyces cerevisiae to form a relatively independent 7-DHC synthesis pathway. At the same time, the storage space of precursor substances required for the synthesis of 7-DHC is increased, and the feedback effect is reduced. In the same compartment, the efficiency of conversion between enzymes is increased, the loss of substrates is reduced, and finally, the yield of 7-DHC is increased by four times, reaching 53.31 mg/L.
