Abstract:
Disclosed in the present invention is a method for preparing pyrrolidone. Provided in the present invention is a method for preparing pyrrolidone from γ-aminobutyric acid under the catalysis of carnitine-CoA ligase CaiC. The amino acid sequence of the carnitine-CoA ligase CaiC is as shown in SEQ ID NO: 1. The ligase has an activity of catalyzing the cyclization of γ-aminobutyric acid to generate pyrrolidone. By means of taking γ-aminobutyric acid as a substrate of the carnitine-CoA ligase provided by the present invention, the yield of pyrrolidone is 3.26 g/L in 24 h, and the molar yield can reach 39.53%, which reduces the production cycle, increases the yield of pyrrolidone, and accelerates an industrialization process for producing pyrrolidone by means of an enzymatic transformation method.
