Abstract:
In order to provide a method whereby an olefin compound can be manufactured with high productivity and to provide an enzyme for use in the method, the inventors introduced variations involving amino acid substitution at various sites of diphosphomevalonate decarboxylase (MVD) and prepared numerous MVD variants. As a result of evaluating the variants for catalytic activity relating to production of an olefin compound such as isoprene, the inventors discovered that MVD in which threonine at position 209 is substituted with another amino acid has catalytic activity, and made it clear that MVD in which arginine at position 74 is substituted with another amino acid has even higher catalytic activity.
