Abstract:

PROBLEM TO BE SOLVED: To obtain a new DNA fragment comprising a DNA fragment having a promoter active part of an alkali protease gene of Acremonium chrysogenum and useful for production, etc., of 7-aminocephem compound for antibiotic raw materials.
SOLUTION: This new DNA fragment has a promoter active part of an alkali protease gene of Acremonium chrysogenum. A 7-aminocephem compound can be obtained in high yield by introducing a vector constructed by binding a gene encoding cephalosporin C acylase represented by amino acid sequence 1-773 represented by the formula into cephalosporin compound-producing bacterium belonging to Acremonium chrysogenum to transform the vector and culturing the bacterium. The DNA fragment is obtained by probing chromosome DNA library of Acremoium chrysogenum with a partial sequence of an alkali protease gene and screening a promoter activity of the resultant gene.
