Abstract:
PROBLEM TO BE SOLVED: To provide a method for efficiently producing 1α, 25-dihydroxyvitamin Dfrom 25-hydroxyvitamin Dunder the presence of vitamin Dhydroxylase, a transformant which expresses this enzyme, and this transformant.SOLUTION: The invention provides vitamin Dhydroxylase having a modified amino acid sequence in which an amino acid at a specific position in a specific amino acid sequence is substituted by other amino acid, and having an activity that under the presence of 0.25 μM of vitamin Dhydroxylase, when reacting with 10 μM of 25-hydroxyvitamin Dand 1 mM of NADPH at 30°C for 20 minutes, the conversion rate to 1α, 25-dihydroxyvitamin Dis 0.30% or more; a transformant which expresses this enzyme; and a production method for 1α, 25-dihydroxyvitamin Dcomprising culturing this transformant under the presence of 25-hydroxyvitamin Dto obtain 1α, 25-dihydroxyvitamin D.SELECTED DRAWING: None
