Abstract:
1389714 Producing cephalosporin C and helvolic acid TAKEDA YAKUHIN KOGYO KK 15 Nov 1972 [15 Nov 1971] 52741/72 Headings C2C and C2U Cephalosporin C and helvolic acid are produced by aerobically cultivating the new microorganism Cephalosporin polyaleurum or a cephalosporin C- and/or helvolic acid-producing mutant or variant thereof in an aqueous nutrient medium and recovering cephalosporin C and/or helvolic acid from the culture broth. The preferred micro-organisms are Cephalosporin polyaleurum 199 (ATCC. 20359, FERM-P No. 1159) and a mutant thereof referred to as Cephalosporin polyaleurum Y-505 (ATCC. 20360, FERMP No. 1160) produced by ultra-violet irradiation of the original organism. Both micro-organisms produce cephalosporin C and hevolic acid together with a small amount of penicillin N. The cultivation is preferably conducted under submerged aerobic conditions in a conventional liquid culture medium. The antibiotics are produced extra-cellularly, and are isolated by filtering off the mycelium, bringing the filtrate to pH 5 and extracting it with ethyl- or butylacetate which takes out the helvolic acid. The aqueous phase is worked up by adsorption on activated carbon, elution with aqueous acetone, concentration of the eluate, absorption on weakly basic ion-exchange resin, elution with ammonium acetate/acetic acid buffer, concentration and after neutralizing with NaOH, precipitating cephalosporin C as the sodium salt by adding acetone. The helvolic acid is obtained from the ethyl- or butylacetate extract of the broth filtrate by concentrating, absorbing on silica gel, eluting with chloroform and evaporating the latter solvent. The examples include for purposes of comparison, a process for producing cephalosporin C by cultivating Cephalosporeum acremonium (ATCC. 14,553).
