Abstract:
Provided are a system for controlling virus replication in response to a hypoxic environment and use thereof. The system comprises: a gene encoding an hypoxia response element, a gene encoding a virus transcription factor recognition element, and one or more genes encoding a virus inhibition factor, wherein said genes are connected in sequence. The system allows the virus inhibition factor, which is a product of a gene of interest, to be expressed in a normal oxygen environment, and to be low-expressed or not expressed in a hypoxic environment, so that the specific killing effect of oncolytic viruses on cancer cells is exerted, thereby improving the curative effect of oncolytic viruses in the treatment of hypoxic diseases such as solid tumors.
