The present invention relates to substituted scleroglucan, a manufacturing method therefor and the use thereof in the manufacture of a drilling fluid composition. The substituted scleroglucan of the present invention has a substituent A, a substituent B and a substituent C respectively or in combination, and optionally has a substituent D and a substituent E respectively or in combination, wherein the substituent A comprises a unit -C(=O)-O- in the structure thereof, and the substituent B comprises a unit -C(=O)-NH- in the structure thereof, the substituent C comprises a unit (I) in the structure thereof, the substituent D comprises an aryl group in the structure thereof, and the substituent E comprises a siloxane group in the structure thereof. The definitions of each substituent are as described in the description. The substituted scleroglucan of the present invention has excellent performances in terms of high temperature resistance, increased viscosity and improved shear strength, as well as a good filtration loss reduction performance and green environmental protection.
