Abstract:
The present invention relates to the technical field of genetic engineering, and provides recombinant Escherichia coli and a construction method therefor, and a method for synthesizing 1,5-pentanediamine. The recombinant Escherichia coli contains an optimal lysine decarboxylase mutant CadAP530L/M569V that catalyzes L-lysine to generate 1,5-pentanediamine, a lysine/cadaverine antiporter CadB that facilitates the entry and exit of a substrate/product into or out of a cell, and pyridoxal kinases I and II that provide self-supply of a cofactor PLP. High-yield 1,5-pentanediamine obtained by using the recombinant Escherichia coli as a catalyst has a high molar conversion rate of 98.67%. According to the method for synthesizing 1,5-pentanediamine provided in the present invention, Escherichia coli is used as a catalyst, no additional cofactor PLP needs to be added, no buffer solution is used (pH does not need to be adjusted), and no hydrochloric acid needs to be used for neutralization, such that the time for synthesizing and converting into 1,5-pentanediamine is short, the production costs are low, and an efficient and economical method is provided for industrial production of 1,5-pentanediamine.
