Abstract:
Disclosed in the present invention is an MMLV reverse transcriptase mutant. Compared with the amino acid sequence of wild-type MMLV reverse transcriptase, the MMLV reverse transcriptase mutant provided in the present invention has mutations at a plurality of sites, for example, site 62, and has the characteristics of improved template conversion performance, improved non-template base addition performance or improved heat resistance. The present invention screens for an MMLV reverse transcriptase mutant having improved conversion performance compared with wild-type templates and further having improved non-template base addition performance or thermal stability. The selected mutant is suitable for RNA sequencing, such as Smart-Seq, nanopore sequencing, and 5'RACE, using a template conversion-based library preparation method, and can also be applied to RT-qPCR and the like.
