Abstract:
According to an aspect, a method for selectively modifying nucleotides comprises a step for reacting nucleotides and a compound of formula 1 in the presence of a rhodium catalyst, wherein the O6 position of a guanine among the four bases of nucleotides can be selectively modified. Specifically, the O6 position of an unpaired guanine can be modified. The reaction can be performed at a low reaction concentration in the presence of a small quantity of catalyst, is kinetically rapid, and can even take place in air. Moreover, all nucleotides, including mononucleotides, oligonucleotides, and polynucleotides, can be selectively modified. Furthermore, by using the method, chemically ligated nucleotides can be produced easily and quickly, photocaged nucleotides, which typically have to go through a complicated synthetic process, can be produced in a single step, a conjugate of a protein and a nucleotide labeled with a fluorescent material can be produced, and even derivatives of nucleotides can be modified, and thus the method can be applied to various techniques utilizing nucleotides containing a modified guanine.
