Abstract:
Disclosed are a columbamine preparation method and a columbamine. The preparation method comprises: taking berberine hydrochloride, adding xylene and trichloroacetic acid to the berberine hydrochloride for reaction at a first predetermined temperature for a predetermined period of time to form a first reactant; adding water into the first reactant, standing for crystallization, converting the crystallized first reactant into hydrochloride by means of a strong base anion exchange resin, and concentrating the hydrochloride to obtain demethyleneberberine; taking the demethyleneberberine, and adding acetonitrile and potassium carbonate to the demethyleneberberine to form a mixture; dropwise adding methyl p-toluenesulfonate to the mixture at a second predetermined temperature, and cooling the obtained mixture to room temperature after the raw materials are completely consumed, to form a second reactant; and concentrating the second reactant, adding potassium hydroxide to the concentrate, performing filtration to obtain a filtrate, adding hydrochloric acid to the filtrate until no precipitate is generated, then using methanol for recrystallization, and finally converting the obtained product by using the strong base anion exchange resin to obtain a columbamine.
